Identify ADH Sequences in Agarose Gel

In humans and many animals, the Alcohol Dehydrogenase (ADH) gene serves to metabolize toxic alcohols in the body. In this experiment, students will identify a simulated ADH polymorphic gene sequence by digestion of plasmids containing the polymorphic genes with restriction enzymes. When the digested samples are analyzed on an agarose gel, students will identify polymorphic genes. Kit includes DNA samples, DNA size ladder, ERCO RI restriction enzyme, restriction enzyme reaction buffer, dilution buffers, agarose, electrophoresis buffers, Instastain® Ethidium Bromide gel stain, and detailed instructions. Requires an electrophoresis apparatus, power supply, automatic micropipette, microwave or hot plate, water bath, and UV transilluminator. Materials supplied for six groups. Time requirements: restriction digestion: 30-60 minutes; electrophoresis: 55 minutes; staining: 15 minutes.
